The performance of the new Kinetex-C18 column was investigated. Packed with a new brand of porous shell particles, this column has an outstanding efficiency. Once corrected for the contribution of the instrument extra column volume, the minimum values of the reduced plate heights for a number of low molecular weight compounds (e.g., anthracene and naphtho[2,3-a]pyrene) were between 1.0 and 1.3, breaking the legendary record set 3 years ago by Halo-C18 packed columns. The liquid-solid mass transfer of proteins (e.g., insulin and lyzozyme) is exceptionally fast on Kinetex-C18 much faster than on the Halo-C18 column. The different contributions of dispersion and mass transfer resistances to the column efficiency were determined and discussed. The possible reasons for this extremely high column efficiency are discussed.  相似文献

Nonlinear propagation of sound has been exploited in the last 15 years in medical ultrasound imaging through tissue harmonic imaging (THI). THI creates an image by filtering the received ultrasound echo around the second harmonic frequency band. This technique produces images of enhanced quality due to reduced body wall reverberation, lower perturbations from off-axis echoes, and multiple scattering of reduced amplitude. In order to optimize the image quality it is essential to be able to predict the amplitude level and spatial distribution of the propagating ultrasound pulse. A method based on the quasi-linear approximation has been developed to quickly provide an estimate of the ultrasound pulse. This method does not need to propagate the pulse stepwise from the source plane to the desired depth; it directly computes a transverse profile at any depth from the definitions of the transducer and the pulse. The computation handles three spatial dimensions which allows for any transducer geometry. A comparison of pulse forms, transverse profiles, as well as axial profiles obtained by this method and state-of-the-art simulators, the KZKTexas code, and Abersim, shows a satisfactory match. The computation time for the quasi-linear method is also smaller than the time required by the other methods.  相似文献

Continuous wave (CW) and transient electron paramagnetic resonance studies have implied that when PsaF is removed genetically, the double reduction of A1A is facile, the lifetime of A1A? is shorter and the ratio of fast to slow kinetic phases increases in PS I complexes isolated with Triton X‐100 (Van der Est, A., A. I. Valieva, Y. E. Kandrashkin, G. Shen, D. A. Bryant and J. H. Golbeck [2004] Biochemistry 43 , 1264–1275). Changes in the lifetimes of A1A? and A1B? are characteristic of mutants involving the quinone binding sites, but changes in the relative amplitudes of A1A? and A1B? are characteristic of mutants involving the primary electron acceptors, A0A and A0B. Here, we measured the fast and slow phases of electron transfer from A1B? and A1A? to FX in psaF and psaE psaF null mutants using time‐resolved CW and pump‐probe optical absorption spectroscopy. The lifetime of the fast kinetic phase was found to be unaltered, but the lifetime of the slow kinetic phase was shorter in the psaF null mutant and even more so in the psaE psaF null mutant. Concomitantly, the amplitude of the fast kinetic phase increased by a factor of 1.8 and 2.0 in the psaF and psaE psaF null mutants, respectively, at the expense of the slow kinetic phase. The change in ratio of the fast to slow kinetic phases is explained as either a redirection of electron transfer through A1B at the expense of A1A, or a shortening of the lifetime of A1A? to become identical to that of A1B?. The constant lifetime and the characteristics of the near‐UV spectrum of the fast kinetic phase favor the former explanation. A unified hypothesis is presented of a displacement of the A‐jk(1) α‐helix and switchback loop, which would weaken the H‐bond from Leu722 to A1A, accounting for the acceleration of the slow kinetic phase, as well as weaken the H‐bond from Tyr696 to A0A, accounting for the bias of electron transfer in favor of the PsaB branch of cofactors.  相似文献

For the first time, large amount of Janus silica particles as small as 100 nm in diameter were prepared through a simple method based on the elaboration of Pickering emulsions of wax-in-water. Controlling the kinetic stabilization of wax droplets allows the fabrication of gram-sized quantities of regular asymmetric inorganic particles with high yield. In fact, our method is based on a limited coalescence process, which allows one to predict the quantity of interface which is produced when working with a known mass of wax, and thus to be sure that all introduced silica particles are adsorbed on the wax surface. To this end, the hydrophilic surface of the silica particles was made partially hydrophobic by adsorbing a known amount of surfactant: cetyltrimethylammonium bromide (CTAB). Varying the concentration in surfactant results in tuning the penetration rate of the particles in the wax droplets, leading to various dimension of the modified area. The broken spherical symmetry of the particle surface was thereafter revealed by the selective adsorption of gold nanoparticles on the amino-modified surface.  相似文献

Ecdysteroids, which are steroid hormones in invertebrates, but which are also present in plants, could potentially be used as anabolic agents in food‐producing animals. The control of ecdysteroid misuse in cattle relies on the development of an efficient method for their detection in biological matrices at trace levels (µg L−1). In order to propose an analytical procedure dedicated to the identification of excreted 20‐hydroxyecdysone (20E) in urine and faecal samples of breeding animals, a comparative study of the spectrometric behaviour of these compounds was carried out both by LC/(ESI‐)/HRMSn (hybrid linear ion trap – orbital trap) and by LC/(ESI+)/MS/MS (triple quadrupole). This study revealed the formation of a large number of product ions both in positive and negative ion mode, corresponding to losses of water molecules and specific cleavages on the side chain. The sample preparation consisted of sequential purification on two solid‐phase extraction cartridges (SPE octadecylsilyl and SPE silica). The detection limits were around 0.5 µg L−1 in the selected reaction monitoring (SRM) mode and recoveries above 60% were obtained. The method was successfully applied to the analysis of real samples collected from calves treated with 60 mg 20E over 4 days. Analysis of the samples allowed the investigation of the kinetics of elimination of 20E in calf urine and determination of the time‐frame for the control of potential abuse. With the recent advances of experimental techniques, the nonlinear ultrafast optical response of metal nano-objects can now be investigated both on ensembles and on single nanoparticles. Its connection with the metal electronic and lattice kinetics is studied on the basis of a model describing the wavelength and time-dependent modifications of the object material dielectric function. Its application is illustrated in the case of single silver nanospheres and gold nanorods, as well as on ensembles of noble metal nanoparticles and metal-semiconductor nano-hybrids. This quantitative analysis also permits to elucidate the physical mechanisms at the origin of ultrafast nonlinearities in confined metals at different timescales.  相似文献

It is shown that exciton swapping between two graphene sheets may occur under specific conditions. A magnetically tunable optical filter is described to demonstrate this new effect. Mathematically, it is shown that two turbostratic graphene layers can be described as a “noncommutative” two-sheeted (2 +?1)-spacetime thanks to a formalism previously introduced for the study of braneworlds in high energy physics. The Hamiltonian of the model contains a coupling term connecting the two layers which is similar to the coupling existing between two braneworlds at a quantum level. In the present case, this term is related to a K-K intervalley coupling. In addition, the experimental observation of this effect could be a way to assess the relevance of some theoretical concepts of the braneworld hypothesis.  相似文献
